This antibody was used to look at changes in tyrosine hydroxylase phosphorylation at serine 31 in mice following MPTP. Seven days post-MPTP brain regions of interest were isolated, homogenized, and purified. Brain samples were evaluated via Western blot.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Brain homogenate

Primary Incubation

1:500 in 5% BSA/TBST. Overnight at 4 degrees

Blocking Agent

5% BSA/TBST

Secondary Incubation

1:10,000 HRP-conjugated secondary, 60 minutes at room temp

Tertiary Incubation

NA

Detection

Femto Chemiluminescent

Results Summary

Using Western blot, we were able to easily detect levels of phospho-TH (ser31) in our brain samples. The binding for this antibody seemed to be very specific and at the correct molecular weight (around 55 kDa).

Additional Notes

Definitely use 5% BSA (dry milk DOES NOT work)

Summary

The Good

Very nice signal

The Bad

Need to use relatively high concentration. Can't use dry milk.

The Bottom Line

If you are looking at phospho-TH in brain homogenate, this antibody works great.
