Extraction of total RNA from brain tissue for use in qPCR experiments. Selected this product in order to purify RNA extracted using phenol: chloroform extraction.
I was examining changes in clock genes in different brain areas via qPCR and needed very pure RNA to start the experiment. I used this kit to extract RNA from tissue and to purify RNA that had been extracted by phenol:chloroform extraction.
Quantitative PCR
Brain tissue in RNAlater or equivalent
Dissect tissue, put in RNAlater. Homogenize tissue in either guanidium isothiocyanate solution or in provided buffer.
Follow all the optional steps!
This kit gave great yields for clean RNA that was intact and suitable for future qPCR analyses. However, using phenol: chloroform extraction gave greater RNA yields in fatty brain tissue than just using the RNEasy kit alone. The phenol: chloroform RNA was not nearly as clean as the RNA from the kit, so both can be combined to get clean RNA at high yield.
I did not examine miRNAs.
Clean RNA, great kit for the cost.
Competitors offer cheaper kits.
This kit is a known good-if you want things to work and be confident that they will do so the first time, this kit is great.
