Fig 2: The percentages of CD4+CD25+CD127+ (inactivated T regulatory cells) in peripheral blood mononuclear cells (PBMNCs) compared between PBMNCs cultured in Quality and Quantity culture media (QQ-media) and PBMNCs cultured in standard culture media. All experiments were performed in triplicate. Forward and side scatter plots were used to gate the lymphocyte populations. The gated cells were then evaluated to identify CD3+CD4+ cells. The CD3+CD4+ cells were then gated to assess the populations of CD4+CD25+and CD127+ cells. Student's t-test was used for phenotypic analysis

Analysis was performed by flow cytometry by using lineage Cocktail-Pacific Blue (Biolegend), CD11c-BV421, CD34-BV421, CD94-BV421, FcERIa-BV421, CD45-VioGreen, CD117-PE (c-Kit), IL7RA-APC (CD127), CRTH2-APC-Cy7 (CD294). ILCs are pregated as CD45 positive cells that are further defined by negative expression of the lineage cocktail and positive staining for IL7RA ( Supplementary Figure 2 ). The figure is presenting cells covering these criteria and their further subdivision by individual expression of surface antigens CD117 and CRTH2. Fig 4: T cell phenotype and senescence/exhaustion markers in MDD(A–D) PBMCs from patients with MDD and matched HC were examined by flow cytometry. CD4+ (upper panel) and CD8+ T cell subpopulations (lower panel) were analyzed for the expression of markers classifying differentiation (CD45RA, CCR7; n = 25) (A), chemokine receptors (CXCR3, CCR6, CCR4; n = 25) or regulatory T cell markers (CD25, CD127; n = 22) (B), markers indicative of exhaustion or senescence (PD-1, KLRG1, CD57; n = 19–20) (C) or inhibitory receptors (CTLA-4, LAG-3, Tim-3; n = 20) (D). Naive/Memory subpopulations were defined as TNaive (CCR7+/CD45RA+), TEM (CCR7-/CD45RA-), TCM (CCR7+/CD45RA-), and TEMRA (CCR7-/CD45RA+). T helper cell subsets were defined as Th1 (CXCR3+/CCR6-), Th2 (CXCR3-/CCR6-/CCR4+), Th17 (CXCR3-/CCR6+/CCR4+), and Th1/Th17 cells (CXCR3+/CCR6+) (n = 25). For the analysis of inhibitory receptors, PBMCs were stimulated with 10 µg/mL of a-CD3 and 1 µg/mL a-CD28 for 48 h. Median and interquartile ranges are shown. Wilcoxon signed-rank test was used to compare groups. All p values > 0.1 if not otherwise indicated.
