A polyphenol oxidase assay typically measures this activity directly by monitoring quinone formation, since the quinone products absorb strongly around 410 nm while their phenolic precursors do not; enzyme activity is calculated from the rate of increase in absorbance at this wavelength as the reaction proceeds.
