Assay Principle

FRAP (Ferric Reducing Antioxidant Power) assay kits measure total antioxidant capacity, a broad indicator of a sample's ability to counteract oxidative damage from reactive oxygen species. A FRAP assay is based on a ferric-to-ferrous reduction reaction: under acidic conditions, a ferric ion-TPTZ (2,4,6-tripyridyl-s-triazine) complex is reduced by antioxidants present in the sample to its ferrous form, producing an intensely colored blue complex with absorbance read in the range of 560 to 593 nm depending on the specific kit formulation. Because this reaction reflects a sample's overall reducing capacity rather than reactivity toward any single free radical species, FRAP is considered complementary to (rather than interchangeable with) radical-scavenging assays such as DPPH or ABTS, and results from the different assay formats do not always correlate directly.
