Alpha-ketoglutarate dehydrogenase (alpha-KGDH, also called the oxoglutarate dehydrogenase complex) is a mitochondrial multi-enzyme complex that converts alpha-ketoglutarate to succinyl-CoA in the TCA cycle, tightly regulated by ATP/ADP and NADH/NAD+ ratios and calcium. An alpha-KGDH assay is generally built around a coupled colorimetric reaction: as the complex converts alpha-ketoglutarate, NAD+ is reduced to NADH, and the electrons are shuttled through a hydrogen carrier to reduce a tetrazolium-based probe (commonly WST-8) to a colored formazan product with absorbance around 450 nm. Enzyme activity is calculated from the rate of color development, referenced against an NADH standard curve or a validated positive control included with the kit.
