Glutathione assay kits are available from several suppliers to determine the levels of glutathione, a major antioxidant in the body. Playing many biological roles, glutathione (GSH) serves as a cofactor for enzymes such as glutathione peroxidase, as a metabolite in the detoxification of xenobiotics, and as an antioxidant compound. The ratio of reduced glutathione (GSH) to oxidized glutathione (GSSG) can be used as a measure of oxidative stress in a cell. In fluorometric glutathione assays kits, orthophthaldehyde (OPA) reacts with GSH, which can be detected by fluorescence. Adding a reducing agent can also convert GSSG to GSH, allowing the total GSSG + GSH content to be determined. To detect GSSG alone, a GSH quencher can be used to remove GSH. Glutathione assay kits may be useful when studying the effects of oxidative stress in conditions such as cancer, diabetes and Alzheimer’s disease.
