The TUNEL assay is a well-established method of detecting cellular DNA fragmentation that occurs from apoptotic signaling. Cells undergoing apoptosis contain damaged or fragmented DNA, which serve as substrates for the enzyme terminal deoxynucleotidyl transferase (TdT). TdT finds DNA double-strand breaks and adds dUTP, which can be modified with labels such as biotin, fluorophores, or antibodies.
