
Caspases, a family of cysteine-aspartic proteases, are protein-degrading enzymes that become activated during programmed cell death, or apoptosis. They play an essential role in ensuring cellular components are degraded without harming surrounding tissue. Assays that measure their enzyme activity, or “caspase assays” have thus become directly tied to investigations of apoptosis. Caspase assay kits and reagents generally work by adding a specific peptide substrate to live cells or cell lysates. Attached to the peptide is a dye that can be detected fluorometrically or colorimetrically upon interaction with the active caspase.
