
Hydrophilic interaction chromatography (HILIC) columns are is used in the separation of polar compounds. HILIC utilizes a polar stationary phase and a mobile phase that is a mixture of water and a less polar organic solvent such as acetonitrile. Hydrophilic partitioning plays the largest role in the mechanism of retention for this method, with other interactions such as ion-exchange, hydrogen bonding, and dipole-dipole contributing to a lesser extent. HILIC is considered a complementary technique to normal and reverse-phase chromatography. Its advantages include enhanced detection sensitivity when used with mass spectrometry, and higher flow rates due to the low viscosity of the mobile phase.
