
Puromycin is an antibiotic that targets protein synthesis. It belongs to the amino nucleoside group of antibiotics and acts by interacting with the large subunit of the ribosome, causing premature termination of peptide chains and inhibiting translation. Puromycin mainly inhibits the growth of Gram-positive bacteria and animal cells, as it has a low permeability in Gram-negative bacteria and fungi. Puromycin resistance is used in molecular biology as a selectable marker, with resistance being conferred by the puromycin acetyltransferase gene (Pac).
