RT-PCR Primers

Primers for RT-PCR (reverse transcription PCR) are available from multiple suppliers. When performing RT-PCR from RNA samples, an initial reverse transcription step takes place to synthesize complementary cDNA. This step requires certain types of primers, such as oligo dT or random primers. Oligo dT (oligo deoxythymidine) primers contain a chain of deoxythymidines, which anneals to the polyA tail of mRNA molecules. Random primers are mixtures of oligonucleotides (usually hexamers or nonamers) that contain random sequences that can be used to direct reverse transcription.
