Sequencing Primers DNA sequencing primers are available from various sources. Among these are primers for regions that flank common sites in cloning and expression vectors. M13 primers target a universal site within the M13 cloning vector, allowing sequencing of cloned inserts. SP6 primers are used in sequencing inserts in vectors containing the SP6 RNA polymerase promoter sequence. T7 primers can sequence inserts downstream of the T7 promoter region of T7 expression vectors. For sequencing mRNA samples, oligo dT primers can be used. Others, such as indexing primers for NGS platforms enable multiplexing when generating sequencing libraries.
