Functionally, this protein is a known histone methyltransferase that dimethylates Lys-36 of histone H3 (H3K36me2). The canonical protein structure is reported to have an amino acid length of 2696 residues, a mass of 296.7 kDa, and is a member of the Class V-like SAM-binding methyltransferase protein superfamily. It is known to be localized in the nucleus and is reported to be expressed in the fetal/adult brain, kidney, skeletal muscle, spleen, and the thymus, and faintly in the lung. As many as 3 protein isoforms have been reported. The NSD1 gene has been associated with the disease, Sotos syndrome.
