Company:

Thermo Fisher

Product Name:

Human Tetraspanin CD63 ELISA Kit

Catalog Number:

EEL135

The CD63 ELISA kit was used to analyze the concentration of CD63 protein in different sets of preparation of exosomes (plasma-derived total exosomes and immunopurified brain-derived exosomes). A dilution of 1:1 was used, and the kit performed well. The gradient of standards was easily seen and the kit was consistent and reliable in those measures. No complaints in general, except for one thing: the wash buffer concentrate (which is supposed to be provided in a vial with 30mL), didn't have any excess into the vial, but just the precise amount - or slightly less than 30mL, which was not supposed to be like that (usually, a small excess come with the vial for automated applications).

Experimental Design and Results Summary

Application

ELISA

Starting Material

Human Plasma; Total Exosomes; and microglia-derived Exosomes

Tips

The kit seems to be suitable for different types of samples, including pure plasma and purified exosomes.

Results Summary

We found that the concentration of exosomes (measured by CD63 levels in pre-prepared samples), differ regarding the method of isolation and/or if the sample was immuno purified.

Summary

The Good

Consistent and good signal-to-noise ratio.

The Bad

Wash buffer concentrate came with a slightly less volume than stated, but didn't affect the results.

The Bottom Line

The kit performed well, and was able to quantify the CD63 concentration from different types of samples/preparations, showing consistency and reliability.
