Worked nicely with RAW cell supernatant, where cells were treated with LPS and ATP for 6 hours. LPS concentration was used 50ng/ml and ATP was used 5mM. We also prepared BMDMs from Wildtype mice. LPS+ATP was used as a positive control to activate the inflammasome.

Follow the protocol correctly.
Inflammasome was activated and the quantifications were optimized with standard curve as it has sufficient range. For 6 hour , 1:50 dilutions were optimum for my experiment.
