Functionally, this protein is believed to provide the missing metabolic reaction required to link the mitochondria and the cytoplasm in the mammalian model of one-carbon folate metabolism complementing thus the enzymatic activities of MTHFD2. The canonical protein structure is reported to have an amino acid length of 978 residues, a mass of 105.8 kDa, and is a member of the Tetrahydrofolate dehydrogenase/cyclohydrolase protein family; Formate--tetrahydrofolate ligase protein family. It is known to be localized in the mitochondria and is notably expressed in most tissues, highest expression found in placenta, thymus and brain. As many as 2 protein isoforms have been reported.
