We were trying to validate GAPDH as an intracellular reference protein in PBMCs from healthy blood donors using flow cytometry. Had some initial trouble with the background, but sorted it out eventually. Overall, happy with the results.

We created a construct for the overexpression of UNC93A and transiently transfected it into HEK293T cells. The FLAG-tagged UNC93A protein was easily detected with an anti-FLAG antibody. However, we were unable to identify a corresponding band using the UNC93A antibody, even at a dilution of 1:100. These results indicate that while the overexpression construct is successfully expressed, the antibody does not recognize UNC93A under the conditions used for Western blotting.
