We work on using stem cells to model and treat rare pediatric diseases. We reprogram patient-derived fibroblasts into iPSCs and utilize this antibody for validation of pluripotency.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

Human iPSCs

Primary Incubation

Diluted 1:200 in IF buffer (1X PBS+ 5% Normal Donkey Serum+0.3% Triton X-100) for 1hr at room temperature

Blocking Agent

IF buffer (1X PBS+ 5% Normal Donkey Serum+0.3% Triton X-100) for 1hr at room temperature

Secondary Incubation

Rb. Cy3 IgG (Jackson Immunoresearch) 1:500 in IF buffer (1X PBS+ 5% Normal Donkey Serum+0.3% Triton X-100) for 1hr at room temperature

Tertiary Incubation

Hoescht 1:10,000 in 1X PBS for 5mins at room temperature

Detection

Imaged on EVOS Fl Auto at 20x magnification

Results Summary

The antibody positively stained the imaged reprogrammed iPSC cluster indicating successful reprogramming and pluripotency. Staining was slightly dim which could be optimized with higher antibody concentration. Imaging was further validated by staining with other pluripotency markers.

Summary

The Good

Affordable with clear cell-specific staining

The Bad

Dim when imaged

The Bottom Line

Overall, a standard Oct4 antibody that demonstrates cell-specific staining for pluripotency validation but is slightly dim on imaging with room for optimization.
