We work on using stem cells to model and treat rare pediatric diseases, especially demyelinating diseases. We differentiate patient-derived iPSCs into neural cells including oligodendrocytes and utilize this antibody as a myelin immunofluorescence marker.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

Human differentiated oligodendrocytes

Primary Incubation

Diluted 1:1000 in IF buffer (1X PBS+ 5% Normal Donkey Serum+0.3% Triton X-100) for 1hr at room temperature

Blocking Agent

IF buffer (1X PBS+ 5% Normal Donkey Serum+0.3% Triton X-100) for 1hr at room temperature

Secondary Incubation

Rt. Cy5 IgG (Jackson Immunoresearch) 1:500 in IF buffer (1X PBS+ 5% Normal Donkey Serum+0.3% Triton X-100) for 1hr at room temperature

Tertiary Incubation

Hoescht 1:10,000 in 1X PBS for 5mins at room temperature

Detection

Imaged on Nikon Spinning Disk at 100x magnification

Results Summary

Patient-derived iPSCs from an ultra-rare demyelinating condition were differentiated into oligodendrocytes. Immunofluorescence with this antibody was used to stain myelin to quantify myelin quantity as well as observe abnormal myelin structure.

Summary

The Good

Clear specific staining, low concentration required

The Bad

Higher cost

The Bottom Line

Overall, a great myelin antibody with low concentration required and consistent staining. The rat host allows for co-staining with various other antibodies.
