We routinely isolate astrocytes from mouse brain tissue in our lab for different downstream applications (Seahorse, Western blot, PCR, RNA-seq, etc.). Myelin isolation is an important step for achieving high-quality astrocyte isolation, ensuring high purity and yield, especially when working with adult mice (we work with mice older than postnatal day 60).

Tips

We work with small tissue samples and add 15 µL of myelin isolation beads to the pellet together with 135 µL of PBS/BSA buffer. The samples are then incubated for 15 minutes before magnetic separation.

Results Summary

We were able to isolate high-purity astrocytes using both magnetic sorting and FACS when myelin was removed beforehand. This allowed us to proceed with the isolated astrocytes for various downstream applications, including RNA-seq, which confirmed the high quality of the isolated material.
