CellMask Deep Red was used to stain the plasma membrane of live primary human brain microvascular endothelial cells (HBMECs).

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

human brain microvascular endothelial cells

Primary Incubation

Different concentrations of working solution were tested (0.5X–1.5X), in HBSS.

Blocking Agent

none

Secondary Incubation

none

Tertiary Incubation

none

Detection

Fluorescent microscopy

Results Summary

We observed a short period of efficient staining with HBMECs using this CellMask. Different working concentrations (0.5×, 1×, and 1.5×) were tested. Among these, the lowest concentration (0.5×) resulted in the least cell death. The internalization was fast (~10 min).

Summary

The Good

Fast method for detection of the plasma membrane in live cells

The Bad

Requires optimization in HBMECs to avoid causing cell death

The Bottom Line

These observations were assessed only in HBMECs, which might be different for other cell types. Nevertheless, it is a fast staining method, at a reasonable price.
