My research focuses on how intestinal immune system matures during breastfeeding phase under the influence of maternal factors. I routinely analyze different immune cell population in mouse gut. Recently I quantified and sorted DCs from mouse gut and I used this Anti mouse IA/IE (MHC2) Pacific Blue antibody from Biolegend to quantify tand sort this cells.

Experimental Design and Results Summary

Applications

ELISA

Sample

Mouse immune cells from any tissue

Primary Incubation

1:3200 at 5-10million cells in 200ul of staining volume

Blocking Agent

Fc blocker

Secondary Incubation

Not needed

Tertiary Incubation

Not needed

Detection

Any flow cytometer with 405 nm laser

Results Summary

The result here is shown for total DC proportion in mouse colonic tissue. The DCs is identified as anti-MHC2 Pac Blue + (Y-axis) and anti-CD11C+ (X-axis). Data is shown for two individual mice. And you can clearly see the staining is very good and bright.

Additional Notes

Titrate the antibody first. MHC2 is highly expressed on myeloid cells, so if you do staining at higher dilution population of interest might go off scale during actual analysis.

Summary

The Good

Extremely Bright, determine the working dilution by titration

The Bad

Though its not negative point, this antibody is very bright so titration is a must, therefore time consuming process.

The Bottom Line

Fantastic signal, I often use this antibody.
