I routinely analyze different T cell population in mouse developing gut. Recently I quantified total T cells subset in mouse gut and I used this Anti mouse TCRb bb515 antibody from Biolegend to quantify the T cells by flow cytometer
Flow Cytometry
Mouse immune cells from any tissue
1:1000 at 5-10million cells in 200ul of staining volume

Any flow cytometer with 488 nm laser
Immune cells were isolated from the small intestine of week 8 mice after collagenase 8 digestion and stained with the fixable viability dye Zombie NIR for 20 minutes on ice. Following viability staining, cells were incubated with BB515-conjugated anti TCRb antibody and stained at a 1:1000 dilution for 30 minutes on ice along with mix of antibody panel. Cells were then fixed and permeabilized using the Invitrogen Foxp3/Transcription Factor Staining Buffer Set according to the manufacturer's protocol, with fixation performed for 30 minutes on ice, Before analyzinf it on BD LSR2. Data here shows total T cells as TCRb+ gate and prior gates are indicated in figure
