The aim of the experiment was to stimulate T cells in a co-culture system of different subsets of T cells isolated from whole blood of humans. The goal of the experiment was to carry out cell suppression assay after T cell stimulation.

Results Summary

We used rIL-2 (200 IU) (PeproTech) and anti-CD3/anti-CD28 dynabeads at 1:1 bead to cell ratio for 5 days at 370C and 5% CO2 in 96 well U bottom plate. In our experiments we successfully stimulate T-cell activation and proliferation in cell culture.

Optimization is needed for the optimal concentration for cell stimulation which is based on the specific cell type being used (e.g., human T cells, mouse splenocytes) and the goals of the experiment.

The Bottom Line

rIL-2 (200 IU) (PeproTech) is very apt to stimulate T cells in cell culture system. For short-term activation (e.g., 24–48 hours) this cytokine can be used in combination with anti-CD3/CD28 antibodies to induce T-cell responses. For 48-hour T cell activation culture, 100 IU/mL IL-2 concentration can be used. However, for long-term culture and higher number of cells increase IL-2 dose to 200 U/mL from day 6 onward.
