Company:

Thermo Fisher Scientific

Product Name:

Dynabeads™ Human T-Activator CD3/CD28 for T Cell Expansion and Activation

Catalog Number:

11161D

The aim of the experiment was to activate and expand T cells in co-culture system consisting of Tregs and CD4+ and CD8+ T-cells separately at 1:2 ratio in 200 μl RPMI supplemented with 5% FBS, rIL-2 (200 IU) (PeproTech) and anti-CD3/anti-CD28 dynabeads (Gibco, Thermo Fisher Scientific).

Experimental Design and Results Summary

Application

Cell culture, Cell stimulation, Cell expansion (Human T cells)

Starting Material

T-cells (Human)

Tips

96 well U bottom plate is needed

Results Summary

The beads with monoclonal antibodies against CD3/CD28 human from Thermo Fisher Scientific specifically reacts with human CD3/CD28 protein and helps in T Cell Expansion and Activation. In our experiments we successfully activated and expand T cells using anti-CD3/anti-CD28 dynabeads (Gibco, Thermo Fisher Scientific at 1:1 bead to cell ratio for 5 days at 37 C and 5% CO2 in 96 well U bottom plate.

Summary

The Good

No need of feeder cells (APCs) and the beads avoids contamination of soluble antibodies or mitogens Easy and efficient activation and expansion/proliferation of T cells (up to 1000 fold)

The Bad

Optimization is needed for the optimal concentration of beads for cell stimulation which is based on the specific cell type being used (e.g., human T cells, mouse splenocytes) and the goals of the experiment.

The Bottom Line

Anti-CD3/anti-CD28 dynabeads (Gibco, Thermo Fisher Scientific, Inc) is very apt to stimulate and expand T cells in cell culture system. The appropriate concentration can vary depending on the specific application, such as short-term activation versus long-term cell expansion.
