I initially prepared a low-range standard curve, but the residual dsDNA concentration in my samples exceeded that range. Using the higher standard curve provided accurate quantification. Protect the PicoGreen reagent from light, incubate the plate for 5 minutes covered with aluminum foil, and then measure fluorescence using a plate reader.

Results Summary

The kit was used to quantify residual dsDNA following IVT mRNA production. After switching to the higher standard curve, the assay accurately measured dsDNA levels with excellent reproducibility. The workflow was simple, requiring only a 5-minute incubation protected from light before reading on a fluorescence plate reader.
