We used a custom BaseScope probe targeting mouse Npas3 mRNA to quantify transcript abundance in astrocytes from adult mouse prefrontal cortex. The probe produced distinct punctate signals with excellent specificity and enabled reliable quantification of Npas3 expression at the single-cell level following conditional gene deletion.

The probe performed exceptionally well when combined with GFAP immunofluorescence and confocal imaging.
Distinct punctate signals were readily detected in astrocytes with minimal nonspecific background. Quantitative analysis clearly distinguished control and conditional knockout animals.
