We are interested in understanding the function of a endososome-localized enzyme and use this anti-Calreticulin antibody as ER marker to validate its subcellular localization not to be in ER. Using sucrose gradient centrifugation and western blotting, we are able to show the enzyme not present in the Calreticulin- fraction.
