Tips

For optimal labeling, I stained exponentially growing Enterococcus cells with 0.5 mM sCy5DL-amide for 15 minutes at 37°C in a shaking incubator.

Results Summary

sCy5DL-amide provided excellent labeling of Enterococcus cells with strong, bright fluorescence and a high signal-to-background ratio. The labeling was consistent across the bacterial population, making it well suited for super-resolution microscopy. From the image, a characteristic tri-septal labeling pattern was clearly visible, indicating active cell wall remodeling at the septa and equatorial regions during cell growth and division. This distinct labeling pattern enabled clear visualization of cell wall dynamics with high spatial resolution.
