My research focuses on vancomycin-resistant Enterococcus faecium (VRE) to understand the molecular mechanisms underlying antibiotic resistance and cell wall biology. As part of this work, I perform splicing by overlap extension (SOE) PCR to assemble DNA fragments into CRISPR-Cas nuclease-encoding plasmids for generating targeted gene knock-ins, and I use the DNA Clean & Concentrator-25 kit to rapidly purify PCR products throughout the cloning workflow.

The DNA Clean & Concentrator-25 kit consistently provided excellent recovery of clean PCR products with minimal sample loss, making it ideal for my SOE PCR cloning workflow. The purified DNA performed reliably in downstream Gibson assembly, restriction cloning, and transformation experiments, enabling efficient construction of CRISPR-Cas plasmids and successful generation of targeted gene knock-ins in Enterococcus faecium.
