I used SuperBlock™ Blocking Buffer for Western blot membrane blocking and antibody incubations. Diluting the buffer 1:1 with TBST produced clean membranes with minimal background, outperforming both milk and BSA in my experiments.

I diluted SuperBlock™ 1:1 with TBST and used the same solution for membrane blocking as well as for diluting both the primary and secondary antibodies. Compared with milk and BSA blocking solutions, this approach consistently produced cleaner blots with lower background

Results Summary

Using SuperBlock™ diluted 1:1 with TBST resulted in strong, specific protein bands with virtually no nonspecific background or dot artifacts after membrane development. The signal-to-noise ratio was consistently better than when using milk or BSA under the same antibody conditions.

SuperBlock™ Blocking Buffer has become my preferred blocking reagent for Western blotting. Using it diluted 1:1 with TBST for blocking and for both primary and secondary antibody dilutions consistently produced clean membranes with excellent signal, minimal background, and no visible dot artifacts, making it superior to milk and BSA in my workflow.
