Company:

Bio-Techne

Product Name:

RNAscope™ Probe - Mm-Cnr1

Catalog Number:

420721

We use RNAscope probe #420721 to measure the expression of Cnr1 mRNA (the Cnr1 gene encodes cannabinoid receptor 1) in mouse brain tissue, for example, the dorsal striatum. This enables us to perform quantitative analysis of cannabinoid receptor 1 expression and compare it between groups exposed to different adverse environmental factors.

Experimental Design and Results Summary

Application

RNAscope in situ hybridization

Starting Material

Mouse brain tissue, fixed frozen sections, 15 µm thick

Tips

We followed the manufacturer’s protocol (“RNAscope Multiplex Fluorescent Reagent Kit v2”, #UM323100) when working with this probe, and for detection we used Opal 650.

Results Summary

Measuring Cnr1 mRNA expression in the dorsal striatal tissue of mice allowed us to assess CB1 receptor expression in mice exposed to cannabis. In the image: dorsal striatal tissue; red—Cnr1 mRNA transcripts; blue—DAPI. Scale bar: 15 µm.

Summary

The Good

Easy to use, produces a robust signal, and worked in all experiments.

The Bad

No bad experience, except a little bit pricey.

The Bottom Line

The probe works very well when the RNAscope protocol is followed. It produces a very robust signal. Definitely recommend it!
