We utilize RNAscope in situ hybridization together with an mCherry probe to visualize the transduced mouse brain region with AAV-GFAP-mCherry. The RNAscope protocol quenches mCherry fluorescence, so additional re-staining is required.

Tips

We used Opal 570 at a dilution of 1:3000 together with the mCherry probe. We followed the procedure described in the user manual (UM 323100) of the RNAscope™ Multiplex Fluorescent Reagent Kit v2.

Results Summary

We were able to visualize the virus-transduced (mCherry-expressing) brain region (CA1 area of the hippocampus). We combined RNAscope in situ hybridization with immunostaining, which allowed us to measure the expression of proteins of interest in the transduced brain region.

Summary

The Good

The probe worked very well, producing a reproducible signal throughout the experiment. It works with a range of mCherry-expressing viruses.

The Bad

No negative experience with this probe. It worked as expected.

The Bottom Line

Highly recommend. Clear, robust, and reproducible signal. The Opal concentration of 1:3000 worked best in our experimental testing (we tested different concentrations) to better visualize the mCherry probe without oversaturation.
