I am investigating how lysosomal damage affects cholesterol transport in lysosomes. I utilize LLoMe because it effectively permeabilizes lysosomes and induces controlled damage to the lysosomal membrane.
Induce lysosomal membrane damage
Live cells
Use at 0.5 to 2 mM dose, for 1 hour treatment.
I observed the formation of Galectin3 puncta on the lysosomal membrane, which is consistent with lysosomal membrane damage induced by LLoMe. This pattern of Gal3 recruitment supports that LLoMe treatment successfully triggered lysosomal permeabilization in my system.
N/A
Cheap, and low side effects.
None.
I observed Gal3 puncta on the lysosomal membrane, confirming LLoMe-induced lysosomal damage.
