In my research, I investigated how a specific cell wall enzyme behaves at different growth stages by measuring its activity and presence through western blotting. I used a dry block heater to pre-heat samples for protein analysis and to conduct in vitro incubations of the enzyme with purified cell wall material at controlled temperatures. This ensured consistent conditions across all experiments. The starting materials included cell pellets and extracted cell wall fractions from cultures harvested at various growth stages, as well as purified enzyme samples. **Results Summary:** The heat block allowed me to maintain precise temperatures for both enzyme incubation and sample preparation, resulting in clear and reproducible differences in enzyme activity and expression patterns across the growth stages, as observed on the western blot. Remarkably, the cell wall hydrolase completely digested the bacterial cell wall after 10 hours of incubation at 37°C using this device.
