The study aimed to quantify the levels of free BDNF in human smooth muscle cells both in the presence and absence of thrombin. To achieve this, growth-arrested cells were treated with thrombin, with and without the PF compound. After two hours of treatment, the cell culture supernatants were analyzed for free BDNF levels using an assay kit from R&D Systems. The selection of this assay kit was based on its effectiveness and supporting literature.

The assay kit was highly sensitive and detected the BNDF levels in picograms. We also found that thrombin has significantly increased free BNDF levels and PF compound blunted these effects.
