Frequent activation of endogenous retrotransposons in cancer cells leads to genomic instability in tumors. To examine the potential anticancer effects of inhibiting retrotransposition, we treated two mouse cancer models (MMTV-HER2/Neu and Th-MYCN mice) that naturally develop breast cancer and neuroblastoma with stavudine, a nucleoside reverse transcriptase inhibitor (NRTI).

Although NRTI was ineffective as cancer prophylactic and had no direct antitumor activity in this MMTV-HER2/Neu model of breast cancer, it caused a dramatic effect on tumor relapses, significantly delaying tumor recurrence and prolonging progression-free survival.
