I am studying the role of BRD4 (Bromodomain-containing protein 4) in regulating gene expression. BRD4 is known for its involvement in various cellular processes, including transcriptional regulation and chromatin remodeling during cancer progression. Acute degradation of BRD4 using d-TAG7 allowed me to study such processes.

Experimental Design and Results Summary

Application

Small molecule degrader

Starting Material

Cells

Tips

Standardize incubation time and concentration based on specific cell lines.

Results Summary

The blot shows expression of BRD4 in IMCD3 cells treated with increasing concentrations of d-Tag7 and incubated for 10 hours. The data clearly shows complete loss of BRD4 under these conditions. I found 50nM d-TAG7 was sufficient for complete degradation of BRD4.
