I've been working with adipocytes for years staining nuclei with Hoescht 33342. Tried out Nucspot Live 650 to see if far red might reduce Hoescht's bleed into the green channel. However, Nucspot Live 650 didn't successfully stain the nuclei when I tried. Their customer support was no help and essentially said "too bad" at the end. We tried over two separate experiments.
Cell Culture Analysis
1X Nucspot Live 650 as indicated in the manual. We've tried Nucspot Live diluted in culture media as well as DPBS.
Doesn't work.
Stained neither the nuclei nor the lipid droplets, seemed to stain something completely different (unidentified). See pic where expected nuclei locations are blank instead of red. (Subsequent experiment with Hoescht showed the correct staining in these locations).
