Company:

Sigma-Aldrich

Product Name:

Anti-RNA polymerase II Antibody

Catalog Number:

05-623

Clone Number:

clone CTD4H8

I study the transcriptional response in response to stress. I have used this antibody for labeling RNA polymerase II in cells.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

IMCD3 Cells

Primary Incubation

1:500 for 1 hr at 4 degrees C

Blocking Agent

BSA

Secondary Incubation

Anti-mouse AF647

Tertiary Incubation

None

Detection

Confocal imaging

Results Summary

The cells show labeling of RNA polymerase II in the nucleus. The antibody has very little background. Also, since this antinody is generated from peptide containing 10 repeats of YSPT[pS]PS corresponding to the carboxyl-terminal domain of RNA polymerase II, the phosphorylated Serine helps to detect active pool of RNA polII during transcription.
