MDA-MB-231 cells underwent nutrient starvation by being cultured in glucose-free DMEM without FBS (Fetal Bovine Serum) for 12 hours, labeled as FBS(-) and Glc(-). The control group was cultured in regular DMEM with 10% FBS, denoted as FBS(+) and Glc(+). Following this pre-culture phase, both groups were subjected to irradiation with doses ranging from 0 to 6 Gy. After irradiation, the cells were maintained in the same culture conditions for an additional 4 hours and then utilized for a colony formation assay. In the Western blot analysis, membranes were incubated overnight with a solution containing 5% BSA in 1xTBST. Subsequently, the secondary antibody was diluted at a ratio of 1:1000, and the membranes were incubated for 3 hours at room temperature.
