Human PBMC were stained with CD3/CD4/CD8, fixed, permeabilized, and then stained with Granzyme B. FIxation and permeabilization using eBioscience™ Foxp3 / Transcription Factor Staining Buffer Set. Antibody was diluted in 1X permeabilization buffer.

Great Granzyme B staining in PBMC but, not monocyte. 1:400 is an ideal concentration to separate positive and negative population. I think one could titrate further. But, it is not essential.
