Functional characterization of T cell. Human PBMCs were stimulated with PMA/ionomycin for 5 hours in the presence of BFA. Cells were then surface stained with CD3/CD4/CD8, fixed, permeabilized, and then stained with IFNg. Fixation and permeabilization using eBioscience™ Foxp3 / Transcription Factor Staining Buffer Set. Antibody was diluted in 1X permeabilization buffer.

Great IFNg staining in PMA-Ionomycin stimulated lymphocyte, but not monocyte. 1:200 could separate positive and negative population. However, would recommend titrated further (i.e. 1:600-800)
