The BV510 CD45 antibody works well for identifying CD45+ immune cells from cell populations isolated from mouse lungs and bronchoalveolar lavage fluid. I found this antibody to provide acceptable separation at 1:500 dilution. However, I recommend the relatively brighter BV570 CD45 antibody from BioLegend which I use at 1:1000.

Results Summary

I used this product to identify CD45+ immune cells from cells isolated from the lung which contains a lot of epithelial and non-immune cell types. Cells were isolated from BALF using 1 ml of 1% BSA in PBS and from lungs through collagenase digestion; RBC lysis was performed using ACK buffer. Cells were then stained with viability dye followed by extracellular antibodies such as CD45, TCR beta, CD19, Ly6C, Gr1. I have not seen any spillover issues with this antibody and fluorophore combination.

BV510 is a dim fluorophore.

The Bottom Line

I would recommend the BV570 CD45 antibody first and if you need to do intravascular staining I would recommend using the BV570 for IV injection and this antibody for the extracellular staining.
