This stain has been used when optimizing flow cytometry-based immunophenotyping of PBMC samples. The results have not yet been published. Multi-colour antibody panels have been designed to identify several lymphocyte populations and tested to ensure appropriate spectral unmixing was possible in a 20+ color panel. This stain enabled the identification of non-viable cells.

Tips

Aliquot unused stain and store at -20oC for future use. Protect from light and avoid repeated freeze/thaw cycles. If using a spectral flow cytometry it is likely that quite significant titration will be required.

Results Summary

Very clear and distinct staining of dead cells. This colour live/dead stain was able to fit into a 20+ colour panel for spectral flow cytometry. Titration to 10% of the manufacturers suggested volume was possible (equivalent of 0.1 uL instead of 1 uL in 1 mL PBS).
