To titrate the antibody, 2 x 10^6 splenocytes from C57BL/6 mice were stained with FITC anti-CD161 Ab and Zombie NIR in 100 µL PBS at 4C for 30 min. Subsequently, the cells were fixed/permed using eBioscience™ Foxp3 / TF Staining Buffer Set according to the manufacturer's instruction, then stained with eF450 anti-EOMES Ab in 100 µL Perm buffer at 4C for 30 min.
