In a previous study conducted in our laboratory, we demonstrated that the presence of multiple copies of mepM, which encodes a paralog of YgeR, compensated for the absence of the D, D-endopeptidase MepS (Singh et al., 2012). Hence, I also checked whether multiple copies of ygeR can compensate for the loss of MepS. The complete get gene was cloned into a pTrc99a vector under the control of an IPTG-inducible promoter (Ptrc::ygeR) and subsequently introduced into the ?mepS strain and checked for the rescue of the mepS knockout phenotype.

Results Summary

A viability assay revealed that the presence of multiple copies of ygeR moderately mitigated the growth deficiency of the ΔmepS mutant on Nutrient Agar media (see figure). These findings suggest that YgeR might exhibit peptidoglycan hydrolase activity.
