We study the role of cell wall hydrolases in the context of bacterial cell wall synthesis. To assess the biochemical activity of these cell wall hydrolases, I overexpressed and purified a protein (MepK) and subsequently isolated it using an affinity chromatography matrix called Ni-NTA matrix from Qiagen.

Include 10-20 mM Imidazole in the lysis buffer to prevent non-specific protein binding.
I successfully purified my cell wall hydrolase, MepK, and subsequently demonstrated its biochemical activity on peptidoglycan. Moreover, I also established that MepK has a role in cell wall synthesis.
