I identified a putative cell wall hydrolase through a genetic screen. After creating a knockout of this gene, I aimed to determine its impact on cell wall synthesis as a pilot experiment. To achieve this, I assessed its antibiotic sensitivity using different cell wall-targeting antibiotics, including cephalexin purchased from Sigma.

Results Summary

A moderate increase in cephalexin sensitivity was observed in the cell wall hydrolase knockout (ΔmepK) when compared to the wild-type (WT) strain, as shown in the figure. However, the antibiotic sensitivity of the ΔmepK mutant was lower when compared to another major cell wall hydrolase mutant (ΔmepS). Nevertheless, the sensitivity increased in the ΔmepS ΔmepK double mutant, as depicted in the figure.
